关于名词性从句和定语从句的接连词的问题

1.在定于从句里面,比如, 1)The branch of science which deals with our lives and health is called biochemistry
2) A great many diseases are caused by germs that get into the body by way of mouth
这是我语法书上的例句, 但是不明白的是第一句用的是which 第二句中没有什么否定代词没有最高级序数词 人加物 等等, 那为什么不选which呢
就像我练习册上的这道 We were all overjoyed at the news__the experiment turned out a success 我选的是which 但答案是that WHY??????

2.还有就是关系代词which的先行词有时是表达主句的概念，老师说就是这的意思,
在联系里我怎么看表达主句的概念啊 不明白啊..

3.最后一个问题 就是名词性从句连接词 what 和 that 的区别..
例 ,___Henry said at he meeting surprised everybody present
我选的是 that 但答案是WHAT..
